Intradigm’s RNAi Nanoplex (NPX) delivery technology offers a uniquely flexible and effective means for the intracellular delivery of active siRNA molecules using systemic delivery. 

Intradigm is leveraging its delivery technology to build a broad pipeline of RNAi-based therapeutic products, known as RNAi NPXs, for the treatment of various serious diseases.
 
In addition to overcoming the delivery challenges in the systemic use of RNAi, the RNAi NPX delivery technology enables a broad therapeutic pipeline through a number of other competitive advantages that can significantly enhance the evolution and efficacy of RNAi therapeutics:

  • Synthetic polymer vectors such as the Intradigm PolyTran™ polymer allow the use of naked (unmodified) siRNA, thus avoiding the need for complicated chemical modifications that may alter the potency and the safety of the siRNA.
  • Intradigm’s proprietary polymers are safe and biodegradable.
  • Self-assembly with the unmodified siRNA based on different electrostatic charge (the positive charge of the cationic polymer attracts the negatively charged siRNA to form nanoparticles). This avoids the use of conjugation chemistry that needs to be individualized for each product.
  • Vectors do not need to be modified with different siRNAs thus enabling a large choice of siRNAs against any target gene for a large number of pipeline and partnership opportunities.
  • Ability to deliver multiple siRNAs directed against multiple genes thus achieving shutdown of several disease targets with one product.
  • PEGylation of the polymers provides the potential to reduce non-specific tissue interactions, increase serum stability and half life, and reduce immunogenicity.
  • Targeting moieties such as RGD ligand can be added, allowing selective distribution to the target tissue.

Intradigm believes that its RNAi NPX delivery technology is one of the most versatile siRNA systemic delivery technologies currently in development for RNAi based therapeutics. 

At present, Intradigm is working to build a high-value internal pipeline of RNAi oncology therapeutics.

Intradigm is working to identify RNAi-based therapeutic products that may effectively treat cancer through silencing several high-value oncology targets. The company possesses an intellectual property portfolio of specific siRNA sequences against more than 50 such targets.  Each of these has the potential to be developed with the RNAi NPX delivery technology as single targeted therapeutics or in combination as multi-targeted therapeutics. 

The flexibility of the RNAi NPX delivery technology provides unprecedented product pipeline opportunities that are difficult to achieve with traditional drug classes such as small molecules or antibodies. 

Examples include:

  • Changes in siRNA “payload” to silence other genes (including resistant mutations) and pathways.
  • Variations of the surface ligand to target different cell types.
  • Variations of the polymer modules to support targeting and intracellular distribution.

Intradigm siRNAs against these targets and many more as single or combination siRNAs in RNAi Nanoplex technology will be used to deliver innovative anti-cancer and anti-inflammatory products.
